Chloroquine then becomes protonated to CQ2+, as the digestive vacuole is known to be acidic pH 4.7; chloroquine then cannot leave by diffusion.

    Its superiority over quinine was established during World War II, when it became the official drug for the treatment of malaria. Quinacrine was introduced for malaria therapy in 1930.
